Protein Function Mitochondrial aconitase (ACO2) is involved in the TCA cycle and catalyzes the isomerization of citrate to isocitrate via cis-aconitate, which associates with ATP-dependent sperm motility.
Biochemical Properties Catalyzes the isomerization of citrate to isocitrate via cis-aconitate. Binds 1 [4Fe-4S] cluster per subunit. Binding of a [3Fe-4S] cluster leads to an inactive enzyme.Highly sensitive to irreversible oxidative inactivation by H2O2.The catalytic residues involved are His-101 and Ser-642.

Significance of PTMs Forms covalent cross-links mediated by transglutaminase TGM2, between a glutamine and the epsilon-amino group of a lysine residue, forming homopolymers and heteropolymers.
PDB ID 1ACO, 1AMI, 1AMJ, 1C96, 1C97, 1FGH, 1NIS, 1NIT, 8ACN,
Additional Comments  low expression of ACO2 might result in low production of isocitrate negatively affecting the TCA cycle to eventually disrupt most sperm motility.
