Secondary Information

Protein Function a potent tumour suppressor gene ,localizes to ER,binds, deubiquitylates, and stabilizes type 3 inositol-1,4,5-trisphosphate receptor (IP3R3), modulating calcium (Ca2+) release from the endoplasmic reticulum into the cytosol and mitochondria, promoting apoptosis.
Biochemical Properties ubiquitin C-terminal hydrolase.Thiol-dependent hydrolysis of ester, thioester, amide, peptide and isopeptide bonds formed by the C-terminal Gly of ubiquitin (a 76-residue protein attached to proteins as an intracellular targeting signal).

DisProt Annotation
TM Helix Prediction No TM helices
Significance of PTMs The ubiquitin-conjugating enzyme UBE2O induces BAP1 sequestration in the cytoplasm by multi-monoubiquitylation of its nuclear localization signal (NLS).
Additional Comments Cells with reduced levels of BAP1 exhibit chromosomal abnormalities and decreased DNA repair by homologous recombination, indicating that BAP1 dosage is critical. Cells with extensive DNA damage should die and not grow into malignancies.
