Protein Function TFII-I is a signal-induced multifunctional transcription factor and it is extremely important for human health and disease.
Biochemical Properties An unusual structural characteristic of TFII-I family members is the presence of a repeated amino acid domain each containing a putative helix-loop-helix HLH-motif characterizing a protein–protein interaction module. TFII-I has six such repeated domains.

Significance of PTMs Tyrosine phosphorylation by Src-family kinase of TFII-I facilitates its nuclear translocation also converts a portion of TFII-I into a transcriptional repressor.Phosphorylation on Tyr-248,377 and 482 contributes to BTK-mediated transcriptional activation.
Additional Comments Gain-of function mutations in the coding regions are found in indolent thymic epithelial tumors (TETs) and Angioimmunoblastic T-cell lymphoma (AITL), suggesting an oncogenic function of TFII-I.
