Biochemical Properties Sp1 contains three zinc finger motifs — as in Zif268, each carries a single α-helix that invades the major groove and makes base-specific contacts. Examination of the structure of a three-finger Cys2-His2 ZFP bound to DNA shows an uninterrupted run of 9 bp that is contacted by the protein viewed edge-on, the protein can be seen as engaging a full 360° of the helix.
